Abstract

[Purpose] To observe the effects of Vitamin K3(VK3) on cell growth and apoptosis in human ovarian cancer cell line SKOV3 and to explore its mechanism.[Methods]The inhibitory effect of VK3 on SKOV3 cells was tested by methyl thiazolyl tetrazolium (MTT) method. The rate of apoptotic cell was quantified by Annexin V/PI assay. The change of survivin gene expression was detected by RTPCR. [Results] The inhibitory rate of VK3(at concentrations of 0,2,5,10,20μmol/L for 48h) on SKOV3 growth was 14.30%, 28.65%, 44.58%, 68.33% respectively. The apoptotic cell rate was 4.36%, 9.87%, 10.80%, 29.65%, 76.80% respectively after SKOV3 cells were treated with VK3 for 48h at concentrations of 0, 2, 5, 10, 20 μmol/L. After treatment with VK3, survivin expression in SKOV3 was downregulated. [Conclusion] VK3 can inhibit the proliferation of SKOV3 cells and induce apoptosis, its mechanism may be related to downregulating survivin gene expression.
