Abstract
Dual-wavelength spectrophotometry was applied to the determination of amylopectin and amylose in the kernel of Castanea henryi through using iodine as coloring agent.On color reaction and photometric optimum conditions were investigated in detail.According to the absorptive spectra of iodine-amylose and iodine-amylopectin,the wavelength of 597nm and 460nm were selected for the determination of amylose,and wavelength of 534nm and 706nm were selected as the measuring wavelength for amylopectin.The content of amylopectin and amylose could be calculated based on the two regressive equations.The recovery rate of both amylopectin and amylose ranged was from 97% to 102%,showed the content of amylopectin and amylose determined by the method was accurate.
