Abstract
OBJECTIVE To explore the relationship between cell apoptosis and Caspase-3 and Caspase-10 protein expression in the hippocampal CA1 of rat with acute cerebral ischemia.METHODS The middle cerebral artery occlusion(MCAO)and reperfusion models were made by intraluminal vascular occlusion in rats.Cell apoptosis was detected by TUNEL method, and the expression of Caspase-3 and Caspase-10 protein was observed by immunohistochemistry and Western-blotting protein imprinting-based respectively.RESULTS ① Apoptotic positive cells were found in the hippocampal CA1 of rat with acute cerebral ischemia after 3h,reached peak after 48h,and reduced after 72h.② Caspase 3 and Caspase-10 protein positive cells were found in the hippocampal CA1 of rat with acute cerebral ischemia after 3h,reached peak after 48h,and reduced after 72h.③ The expression of Caspase 3 and Caspase-10 were positively correlated with the numbers of apoptotic neurons in the hippocampal CA1 of rat(r = 0.976,P﹤0.01;r = 0.986,P﹤0.01).CONCLUSION The hippocampal CA1 of rat with acute cerebral ischemia existed apoptosis, and it was consistent with the expression of Caspase3 and Caspase-10 protein.Caspase 3 and Caspase-10 protein may promote apoptosis.
