Comparative study on the functional recover of axoplasmic transport of regenerative nerve promoted by nerve growth factor or ciliary neurotrophic factor

Abstract

Objective:To compare the effects of recover of axoplasmic transport of regenerative nerve promoted by nerve growth factor(NGF) or ciliary neurotrophic factor(CNTF).Method:Thirty-two Sprague-Dawley(SD) rats with 10mm defect of sciatic nerve were randomly divided into two groups which had been bridged with the new designed double channel nerve conduit of fusiform shape. Each groups contained twelve animals, in the first group, 200μl of chitin for medical use was injected into the conduit, in the secondgroup, the two branches of the conduit contained 100μl of the chitin and 5 μl NGF or CNTF.At the 12~(th) and 16~(th) week after operation,the authors first evaluated the axoplasmic transport of regenerative nerve in the two branch channels with Horseradish peroxidase(HRP) and Nuclear yellow(NY),and observed quality and quantity of the regenerative fibres with Holmes staining.Result:At the 12~(th) and 16~(th) week after operation,there were not significant differences of the regenerative nerve fibres between two channels in the first groups, but in the second group, there were more and well-arranged regenerative fibres in branch contained the CNTF, when the tracing substance of (HRP) and NY were injected to the regenerative nerve in the branch contained CNTF, the positive neurons in the spinal cord was much more and densely distributed.Conclusion:The effects of CNTF on functional recover of axoplasmic transport of regenerative fibres is superior to that of NGF
