Abstract

Objective To observe the changes of Bcl-2 and Bax protein expressions in rat retina after acute injury and study the intervention effects of Huoxuehuayutang on Bcl-2 and Bax protein expressions.Methods Sixty-five rats were randomly divided into control group(A),pressure-loading group(B) and pressure loading-Huoxuehuayutang treatment group(C).After acute injury,the Bcl-2 and Bax protein expressions at different time points were detected by immunohistochemical method.Results The Bcl-2 and Bax protein expressions were seen in the retinal ganglion cell layer in Group A.In Group B,the Bcl-2 protein expression began increasing at 2 hr,eached the peak at 36 h,recovered to normal at 72 h.Compared with Group A,Bcl-2 and Bax proteins in Group B significantly increased from 12 h to 48 h after acute injury(P0.01.) The Bax protein expression was similar to that of Bcl-2 protein except that the peak appeared at 24 h.In Group C,the Bcl-2 protein expression significantly increased from 12 h to 48 h after acute injury compared with Group B(P0.05),but there was no statistical difference in Bax protein expression between two groups(P0.05).Conclusion The Bcl-2 and Bax protein expressions increase in rat retina after acute injury.Huoxuehuayutang can up-regulate the Bcl-2 protein expression,but has no effect on the Bax protein expression.
