Effect of rosiglitazone on NO and ICAM-1 secreted by HUVEC under high glucose

Abstract

Objective To investigate the effect of high glucose on excretion of NO and ICAM-1 in human umbilical vein endothelial cells at different times,and to explore the intervention effect of rosiglitazone.Methods Human umbilical vein endothelial cells were cultured in DMEM medium with or without high glucose for different times(0 h,24 h,48 h,72 h,96 h).The cells were randomly divided into three groups:control group,high glucose group and rosiglitazone group.NO level was determined by detecting NO-3+NO-2 in supernatant,with HNO3 reductance assay.Meanwhile ICAM-1 in supernatant was detected by ABC-ELISA.Results Compared with control group,the concentration of NO was significantly increased at 24 h and then decreased as cultured time lengthened in high glucose group.It was significantly decreased at 48 h and then down to base line at 72 h.Meanwhile ICAM-1 level was increased time-dependently.Compared with control group,rosiglitazone could inhibit the effect of high glucose on NO and ICAM-1 in rosiglitazone group.Conclusion Rosiglitazone can correct the endothelial cell dysfunction induced by high glucose and protect the vascular function.
