Abstract

Objective To study characteristics of injury in apoptosis of HL\|60 cells and its related genes bcl\|2 and bax expression induced by enriched uranium 235 U. Methods DNA gel electrophoresis was used to show the apoptosis of HL\|60 cells.Immunohistochemical technique was used to identity the expression of bcl\|2 and bax proteins in HL\|60 cells,meanwhile RNA dot blot hybridization was used to show the expression of bcl\|2 mRNA in HL\|60 cells. Results HL\|60 cells irradiated with 235 U displayed characteristics of DNA ladder formation in apoptotic cells.While the expression of bcl\|2 protein was as high as (88±7)% in unirradiated control cells,down\|regulation [lowered to (61±5)%] of its expression was noted after exposure to 235 U.The expression of bax protein was low in control cells,and after irradiation it did not change obviously.A marked down\|regulation of bcl\|2 mRNA expression was found in irradiated HL\|60 cells. Conclusion Progression of apoptosis in HL\|60 cells induced by 235 U is dependent on the time elapse of 235 U irradiation.The apoptotic action of 235 U in HL\|60 cells is associated with down\|regulation of the apoptosis\|related gene bcl\|2.\;
