Abstract

Objective To investigate the inhibitory effects of mediated PTEN gene in combina- tion with L-OHP on human cholangiocarcinoma cell line.Methods A eukaryotic expression vector containing PTEN gene was transfected into human cholangiocarcinoma cell line QBC-939 under media- tion of lipofectamine,and the positive cell clones were selected and amplified.The expression of PTEN gene was determined with immunohistochemistry.Cell growth curve was illustrated and MTT performed to test cell invasion.Electron microscopy was conducted to determine the effects of PTEN on cell cycle,uhrastructure and cell growth of the transfected QBC-939 cells and on tumorgenicity in nude mice.Results PTEN was significantly expressed in the transfected QBC-939 cells(P0.05). The cell growth was slower than in other groups(P0.05).The size of transfected cells changed. The growth of the transfeeted cells was inhibited at phase G1 to S and its apoptotic rate markedly in- creased(P0.05).The cell growth in nude mice was inhibited after addition of L-OHP.Conclusions The inhibitory effects of PTEN on eholangiocarcinoma cell line can be strengthened by L-OHP.
