Abstract

Objective To Evaluate the effect of celecoxib,a selective inhibitor of cyclooxygenase-2 (COX-2) on proliferation,moving and invasiveness of glioma U251 cells.Methods The inhibition rate of U251 cells was growth determined by methyl thiazolyl tetrazolium (MTT) assay 12,24,36 and 48 hours after the treatment of U251 cell with celecoxib of different concentration.The expression of COX-2 protein in U251 cells was determined by a laser focusing microscope 12 and 24 hours after the treatment with celecoxib.The expression of proliferating cell nuclear antigen (PCNA) in U251 cells was determined by immunohistochemical technique 24 and 48 hours after the treatment with celecoxib.The inhibitive effects of celecoxib on U251 cells moving and invasiveness were determined by transwell technique.Results The inhibition rate of U251 growth significantly increased with increase in the concentration of celecoxib and time of treatment (P 0.01).The expressions of COX-2 protein and PCNA in U251 cells decreased significantly after the treatment with celecoxib of concentration of 100μmol/L compared to those in the control group (P 0.01).The moving and invasiveness of U251 cells were significantly inhibited by celecoxib.Conclusion The growth,proliferation,moving,expression of COX-2 protein and invasiveness of U251 cells may be inhibited by celecoxib in dose-and time-dependent manner.
