Abstract

Objective To explore the protective effect of serum contained Astragalus membranaceus on endothelial cell and mechanism. Methods Human umbilical vein endothelial cells(HUVECs) ECV-304 cultured in vitro were divided into the negative control group,the control group and the serum contained Astragalus membranaceus intervention group.Endothelial cell damage mode was established by using angiotensin Ⅱ(Ang Ⅱ),and serum contained Astragalus membranaceus were used to intervene.Flow cytometry was used to test the apoptosis rate of endothelial cell and immunoblotting was used to test the expression of heat shock protein 70(HSP70) in endothelial cell. Results Ang Ⅱcould induce endothelial cell apoptosis.Different concentration of serum contained Astragalus membranaceus were significantly reduced the apoptosis rate of endothelial cells induced by AngⅡ(P0.05),and could obviously promote the endothelial cells of HSP70 expression(P0.05). Conclusion Serum contained Astragalus membranaceus can inhibit endothelial cell apoptosis that is induced by Ang Ⅱ,and promoting the expression of HSP70 may be one of the mechanism.
