Abstract
Objective To analyze the correlation among blood glucose,insulin and injured pancreatic islet cells in rats with type Ⅱ diabetes,to supply some evidence for investigating islet dysfunction in type Ⅱ diabetes and for develoming anti-diabetic drug.Methods Male Sprague-Dawley rats were given one dose of streptozotocin(STZ,30 mg·kg-1) by intraperitoneal injection after feeding with high fat and sugar diet for 5 weeks.Three days later,rats were tested for 4-hour fasting blood glucose(4hBG),and 4hBG level between 11.0 mmol·L-1 to 25.0 mmol·L-1 was chosen as the standard for a successful model.The rats in the model group and high fat and glucose groups were fed with high fat and sugar diet for 4 weeks,and then the 4hBG,serum insulin level and pathological changes of pancreatic islet were objectively recorded.Results Compared with the control group,the blood glucose level in the model group was significantly increased 72 hours after injection of STZ,and remained high level till the end of the experiment.At the same time,the β cell function index decreased obviously,and islet cells were seriously injured.Conclusion After fed with high fat and sugar diet and intraperitoneal injection of low-dose STZ,blood glucose levels and insulin secretion of rats are increased,islet function is severely impaired,showing that the damage of pancreatic islet cells is positively correlated with blood glucose and insulin secretion.
