Abstract
Objective To study the optimum extraction technology and antioxidant activity of polysaccharides from the fungus #CSL-8,firstly isolated from Crocus Sativus L.Methods The extraction of the polysaccharides was prepared by L9(33) orthogonal test plan,the antioxidant activity of the extract was evaluated by DPPH method,compared with Vc.Results The optimal extraction condition of fungal polysaccharide in water extraction was as follows: extraction temperature 95℃,material-water ratio 1: 30,and extraction time 4h,the rate of extraction was 9.47%,the concentration of polysaccharide in the fungus was 26.35%.The optimal extraction condition of fungal polysaccharide in ultrasonic extraction was as follows: extraction power 400w,material-water ratio 1: 40,and extraction time 40min,the rate of extraction was 9.12%,the concentration of polysaccharide in the fungus was 57.65%.In a certain concentration scope,there was a strong positive correlation between the scavenging activity and the concentrations.The water extraction had an IC50 value of 0.15 mg/ml,and the ultrasonic extraction was 0.11mg/ml.Conclusion The ultrasonic extraction surpassed the water extraction.The tested polysaccharides by two extraction methods has strong ability to scavenge the stable free radical DPPH and shows a concentration-effect relationship.
