Determination of pantoprazole in human plasma by HPLC-MS

Abstract

OBJECTIVE To develop a HPLC-MS method for determination of pantoprazole in human plasma.METHODS The method was performed on Waters Atlantis~(TM) C_(18) column, (2.1mm×100mm,3.0μm) at 30℃, 0.1% formic acid: acetonitrile (40∶60) was used as mobile phase with the flow rate of 0.2ml·min~(-1), the injection volume was 5ul. Electrospray ionization(ESI) source was applied and operated in the positive ion mode. Selected reaction monitoring(SRM) mode was used to quantify pantoprazole. The parent ion of pantoprazole and internal standard paroxetine \[M+H\] m/z were 384.2 and 330.1, respectively, while the product ion for pantoprazole and internal standard were 200.5 and 192.1, respectively. RESULTS The assay was linear over the range of 10~5000ng·ml~(-1), the limit of detection was 1.0ng·ml~(-1), the intra-day and inter-day precision (RSD) ranged from 1.8 to 4.0%, and 1.2 to 1.7%, respectively, at three different concentrations. CONCLUSION The method developed is simple, sensitive and fast. It can be applied to determine pantoprazole in human plasma.
