OBJECTIVE:To establish the SYBR Green I Q-RT-PCR method for detection of the quantification of hMAM mRNA as the quantification marker of CTCs in peripheral blood of breast cancer patients and to investigate its feasibility for diagnosis of the micrometastases of breast cancer cells into peripheral blood and its significance.METHODS:SYBR Green I Q-RT-PCR was established and applied to the quantitative detection of hMAM mRNA in peripheral blood samples from 61 patients with breast cancer and 33 patients with benign breast tumor and 11 healthy volunteers.The correlations between hMAM mRNA expression in the peripheral blood of breast cancer patients and patients' clinical and pathological features were analyzed.RESULTS:hMAM mRNA was expressed in 16.4%(10/61)of peripheral blood of breast cancer patients:8 cases were HER-2 overexpression in primary tumors,the cases of HER-2-neg ative shown no hMAM mRNA expression,and the difference was significant statistically(P=0.028),and there were no significant differences with other clinical and pathological features(P0.05).The quantification of hMAM mRNA in peripheral blood of breast cancer patients were in range of 1.98×102-6.21×103(median 4.0×102).There was no significant difference between the quantification of hMAM mRNA and patients' clinical and pathological features(P0.05).hMAM mRNA was negative in peripheral blood of the benign breast tumor patients and healthy volunteers.CONCLUSIONS:The SYBR Green I Q-RT-PCR method can be used to detect the quantification of hMAM mRNA in peripheral blood of breast cancer patients as a marker for diagnosis of micrometastases of the breast cancer cells into peripheral blood.The hMAM mRNA expression in peripheral blood of breast cancer patients is significantly related to the HER-2 status of primary tumor.
