THE EFFECTS OF PROTEIN KINASE C IN TGF-β_1 STIMULATING PROLIFERATION AND COLLAGEN SYNTHESIS OF NORMAL AND HYPERTROPHIC SCAR FIBROBLASTS

Abstract

Objective:To investigate the effects of TGF -β 1 on activity of protein kinase C(PKC),proliferation and collagen synthesis of fibroblast derived from hypertrophic scar and normal skin(HS -FB and NS -FB).Methods:The PKC activity was detected by 32 P incorporation assay.Cell proliferation was measured by methyltetrazo lium assay.Collagen synthesis was measured by 3 H -proline in-corporation assay.Results:The PKC activity of NS -FB and HS -FB wa s enhanced by TGF -β 1 .TGF -β 1 could increase markedly proliferation and collagen synthesis of the two types of FB(P0.05).PKC activator(phosphatidyl serine,diacylglycerol and Ca 2+ )had also same effects(P0.05)while GF109could abrogate partly ro les of TGF -β 1 (P0.05).Conclusion:TGF -β 1 could activate PKC and could stimulate proliferati on and collagen synthesis of the two t ypes of FB,but PKC inhibitor could ab rogate partly these roles.It might indicate that the stimulato ry effects of TGF -β1might be partly mediated by the PKC pathway.[
