Abstract
Cotyledon and hypocotyl of buckwheat were inoculated and cocultured with Agrobacterium rhizogenes A 4 strain .The transformed hairy roots was effected by different culture conditions.The hairy roots in liquid medium grew more faster than that in solid medium.The medium inculuding certain concentrated Cu 2+ could promote the growth of hairy roots.Different explants and preculturing time showed certain influences to the transformation .The transformation frequency of hypocotyl was higher than that of cotyledon. Before the inoculation with Agrobacterium , a two day preculture of recipient explants on the phytohormone containing medium could obviously promote the transformation.Paper electrophoresis revealed that the hairy roots synthesized agropine and mannopine.
