Adenovirus vector expressing p16 gene suppresses the growth of human glioma cell line

Abstract

Objective To study the effect of the adenoviral vector(Ad5)expressing p16 gene on the growth of human glioma cell lines TJ899 and TJ905.Methods The DNA of adenovirus was extracted and amplified,then innoculated into 293 cells.Ad-p16 was transfected into TJ899 and TJ905 cells at multiplicity of infection(MOI).The expression of p16 was detected in TJ899 and TJ905 cells by immunohistochemical method.The proliferation of TJ899 and TJ905 cells was analyzed by methly thiazolyl tetrazolium(MTT)and colony formation.Results The p16 expression was observed in TJ899 and TJ905 cells.The growth rate of the Ad-p16 infected TJ899 and TJ905 cells was inhibited by 93% and 98.3%,as compared with that of the adenovirus with β-lactase gene(Ad-LacZ) infected cells.Colony formation in the Ad-p16 infected cells remarkably decreased as compared with Ad-LacZ infected cells.Conclusion The exogenous p16 mediated by adenovirus can inhibit the growth and colony formation of glioma cells.
