Determination of Ractopamine in Swine Urine by Gas Chromatography-Mass Spectrometry

Abstract

A gas chromatography-mass spectrometric (GC-MS) method has been established for the determination of ractopamine in swine urine.The samples were hydrolyzed by β-glucuronidase/arylsulfatase, extracted with ethyl acetate and hydrochloric acid solution,cleaned up on SCX cartridge, derived with N,O-bis(trimethylsily1) trifluoroacetamide(BSTFA), and detected by GC-MS.The results indicated that the limit of detection (LOD) of ractopamine in swine urine was 0.3 μg/L, the limit of quantification (LOQ) was 0.5 μg/L,and the linearity was within the range of 10~500 μg/L.The mean recoveries of swine urine samples fortified at three levels of 2, 4 and 8 μg/L was within the range of 85%~115%, and intra-and inter-batch relative standard deviation were less than 10%.
