Abstract

The development of a new rabies vaccine prepared from CTN strain virus propagated in primary hamster kidney cell (PHKC) culture was performed. The virus seed of CTN was serially passaged in PHKC for adaptation and the rabies vaccine was prepared by using infected cell culture as virus seed.After more than tenth passages the CTN virus was adapted in PHKC with a virus titer around 7.0LogLD 50/ml. The potencies of three lots of purified vaccine made by the adapted virus (CTN-LS-HK)were 6.11~6.55IU/ml that appeared higher than the vaccines made by aG strain of rabies seed virus grown in guinea-pigs brain (3.77~5.85 IU/ml). The CTN adapted virus strain can be used for production of a more immunogenic and safer rabies vaccine, which doesn't contain brain tissue and will avoid the contamination of extraneous agents in the guinea-pigs.
