Study of the pathology changes influenced on small dose of NOS inhibitor in the focal cerebral ischemia

Abstract

Objective To study the relation of nitric oxide (NO) and pathology changes in the focal cerebral ischemia. Methods Using of light microscope and electriomicroscope and TUNEL method the pathological changes were observed in the focal cerebral ischemia. Results In the 1d of focal cerebral ischemia and reperfusion for under light microscope,neuronal cell were necrostic in cortex region and hippocampus. Under electriomicroscope there were only chromatin dense stain (likely an apoptosis body) of neuronal cell nucleus and distortion of gliocyte nucleus in ischemic penumbra in the operation side of brain. They showed apoptosis. But the pathology changes of necrosis and apoptosis under the influence of nitric oxide synthase (NOS) inhibitor [ N nitro L arginine (NNLA)] were lighter than those in the focal cerebral ischemia. TUNEL results showed:The change of NO content influenced percent rate of apoptosis cells of DAB positive stain in infarction. Conclusion In the process of focal cerebral ischemia and reperfusion,regulating the quantity of NO in the propriety range may protect and avoid purpose of neuronal cell injury.
