Abstract

Objective To observe the effect of arsenic trioxide(As2O3)on proliferation and expression of collagen type I of 3T3 fibroblasts induced by recombinant interleukin 13(rIL-13). Methods 3T3 fibroblasts were cultured in vitro,which were divided into four groups:DMEM control group,rIL-13(80 ng/mL)group and rIL-13 with various concentrations of As2O3(2 μmol/L and 4 μmol/L)group.MTT assay was carried out to study cell proliferation;collagen type I(CoI I)was determined with Western blot. Results As2O3 significantly inhibited the proliferation of 3T3 fibroblasts induced by rIL-13 with a dose dependent manner(P0.01).The expression of collagen type I could be detected in all groups,Which in rIL-13(80 ng/mL)group was the strongest among four groups.But the expression of collagen type I in rIL-13 with different concentrations of As2O3(2 μmol/L and 4 μmol/L)groups was significantly lower than that of rIL-13 group(P0.01),and there was no significant difference between 4 μmol/L As2O3 + rIL-13 group and 2 μmol/L As2O3 + rIL-13 group(P0.05). Conclusion As2O3 may play an important role in inhibiting the proliferation and expression of collagen type I induced by interleukin-13.
