Construction and expression of human PSCA gene eukaryotic expression vector

Abstract

Objective To construct the eukaryotic expression vector for human PSCA gene and express it in vitro. Methods Total RNA was extracted from LNCaP cells and the DNA encoding entire human PSCA gene was amplified by RT-PCR,and then inserted into eukaryotic expression vetor pcDNA3.1(+) plasmid. The recombinant plasmid pcDNA3.1(+)-PSCA was transfected into Hela cells and the expressed product was detected by RT-PCR and immunocytochemistry. Results RT-PCR and immunocytochemistry demonstrated the exist of PSCA in the recombinant plasmid-transfected Hela cells. Conclusion The constructed DNA vaccine can be expressed in vitro,which may pave a way for further studies in animals.
