Effects of benazepril on apoptosis and expression of Bax and Bcl-2 in the kidney of diabetic rats

Abstract

Objective To investigate the effects of Angiotensin converting enzyme inhibitor on apoptosis and the expression of Bax and Bcl 2 in the kidney of diabetic rats. Methods Diabetes was induced in uninephrectomized SD rats by intraperitoneal injection of STZ(65 mg/kg). From the next day benazepril(10 mg/kg) was delivered daily by gavage for 12 weeks. Apoptotic cells in renal cortex were detected by means of terminal deoxynucleotidyl transferase mediated d UTP nick end labeling (TUNEL). Flow cytometry was used to evaluate the expression of Bax and Bcl 2. Results Compared with those in the kidneys of control group, apoptotic cells were more in number and the expression of Bax and Bcl 2 was higher in the diabetic kidneys (P 0.05 ), the increase of Bax/Bcl 2 ratio was also observed (P 0.05 ). By benazepril treatment, the number of apoptotic cells and the level of expression of Bax and the Bax/Bcl 2 ratio decreased, the expression of Bcl 2 increased(P 0.05 ). Conclusion The renal protective effect of angiotensin converting enzyme inhibitor benazepril on diabetic kidney may be partly through inhibiting apoptosis by regulating Bax and Bcl 2 expression.
