Influence of As_2O_3 on human glioblastoma cell line U251 in vitro

Abstract

Objective It is to discuss the action and mechanism of As_2O_3 on human glioblastoma cell line U251 cultivated in vitro. Methods The survivorship, morphology change, distribution of cell DNA content of U251 cells in the groups given medicine of different concentration and control group were observed and measured with inverted phase contrast microscope, MTT method and flow cytometry. Results The concentration of 0.5~4 μmol/L of As_2O_3 could obviously inhibit the proliferation of U251. Flow cytometry showed that the peak of hypodiploid occurred before G1 stage in every group given medicine, and cells decreased with apoptosis of induction cells in G_0/G_1 stage. Conclusion As_2O_3 can inhibit the proliferation of human glioblastoma cell line U251 in vitro. It can be used to treat glioma.
