Abstract
【Objective】 To study the content,activity and significance of the splenic dendritic cells(DCs) of severely scalded mouse.【Methods】 To replicate the 30% scalded mouse experimental model,the mouse were divided into normal control group and experimental group,and the experimental group was divided into 4 h,12 h,24 h,48 h,72 h group,10 mouse in each group.The expression of CD11c+ and co-stimulatory molecule CD86 on splenic dendritic cells were examined by immunofluorescence labeling,and the content on CD11c+ splenic dendritic cells were examined by flow cytometry.【Results】 In the normal control group,there is only a small amount of CD11c+ DCs in the spleen.In the 4~12 h experimental group,the numbers of CD11c+ DCs and CD86+ DCs in spleen were significantly increased compared with normal control group(P 0.05).From the 24 h experimental group,the content of DCs started to decrease,especially the content of CD86+ DCs which reflects the DCs Activity was significantly decreased compared with the control group(P 0.05).【Conclusion】 Shortly after the burn,spleen DCs had a transient increase in the numbers and activity,then the numbers of DCs was significantly reduced,and the activity was significantly decreased,which suggested that the changes of spleen DCs after burn were closely related with the cellular immune function.
