Effect of tea polyphenols on proliferation of human umbilical vein endothelial cells

Abstract

Objective To study the effect of tea polyphenols(TP) on the proliferation of human umbilical vein endothelial cells(HUVEC) in vitro by the variance analysis of factorial design.Methods The effects of TP on the proliferation of HUVEC were detected by two-factor 3×5 factorial design and methyl thiazolyl tetrazolium(MTT) assay in different concentrations and different time periods.The morphological changes of HUVEC were observed by using the inverted phase contrast microscope.Results In 24-hour period the groups with TP concentrations of 120,160 and 240 mg/L,and in 48-hour and 72-hour periods the groups with TP concentrations of 80,120,160 and 240 mg/L had significantly different OD values compared with those of the control group.The values of IC50 were 184.41,127.70 and 108.98 mg/L on 24th,48th and 72nd hour respectively.There was an interaction between effective period and concentration of TP.Conclusion Tea polyphenols can inhibit the proliferation of HUVEC in vitro,and there is a dependent relationship of dose-effect to time-effect.
