Abstract
Objective To determine whether arctiin from Fructus Arctii could affect the growth of C2C12 myoblasts and the differentiated myocytes and identify its mechanism.Method The growth of C2C12 myoblasts and the different-tiated myocytes induced by 2% horse serum was observed in vitro by treatment with different doses of of arctiin(0,0.01,0.1,1,10,100 and 1000 ng/ml).Daidzein was used as control.C2C12 myoblasts' proliferation was assessed by MTT method,and the differentiated myoblasts' total protein,CK activities and the expression of estrogen receptor β(ER β) were detected by Coomassie brilliant blue method,colorimetric analysis and QRT-PCR,respectively.Results C2C12 myoblasts' proliferation was significantly upregulated(P0.01).The differentiated myocytes' total protein(P0.01) and CK activities(P0.01) were markedly increased after treatment of actiin at the concentration of 10ng/ml.Meanwhile,the expression of estrogen receptor β(ER β) in differentiated myoblasts' was increased by actiin at the same concentration.Conclusion Arctiin can facilitate the proliferation of C2C12 myoblasts,and the hypertrophic growth of skeletal muscle cell after differentiation by binding to ERβ located within C2C12 myoblasts and differentiated myocytes.Consequently,arctiin might become potentially a new type of green and effective physiological modulator.
