Effect of Recombinant Adenovirus-mediated Wild-type p53 Gene Transfection on Lymphoma Cells

Abstract

Objective The purpose is to observe the expression of the P53 protein and its effects after the transfection of recombinant adenovirus-mediated wild-type p53 gene drug and the joint radiation on the lymphoma cell lines(raji and daudi).Methods To determine the inhibition rates of Raji and daudi cells interfered with different concentrations of drug,different transfection time,different radiation dose and the joint radiotherapy treatment with MTT assay.Daudi and raji cells were divided into the control group,drug group,radiotherapy group and joint group.Western Blotting assay was used for detecting the protein after the extraction of each group and the images were scanned for analysis.Spss14.0 software was used for single-factor analysis of variance.Results The inhibition rates of the drug groups of raji and daudi cells were significantly higher than that of the control group(P0.05).Inhibition rates increase with the radiation dose in radiotherapy groups of two cell lines.Inhibition rate of daudi cells in the joint group of each dose point was not significantly different from the radiotherapy group(P0.05).P53 protein expressions of the both experimental groups showed slightly higher than that of the control group The protein expressions of the joint groups of raji cells and daudi cells have no significant difference with the radiotherapy group(P0.05).Conclusion The adenovirus-mediated p53 gene drug can transfect the lymphoma cells,and express the P53 protein,the inhibitory effects for raji and daudi lymphoma cells are to a certain extent.Recombinant adenovirus-mediated p53 gene drug can't increase the effect of radiotherapy.
