Abstract
OBJECTIVETo study the effect of ectogenic IL-10 on expression of ICAM-1 in hepatic stellate cells of hepatic fibrostic rat. METHODS60 male SD rats were divided into three groups randomly: 8 in normal control group (group N), 28 in carbon tetrachloride group (group C), and 24 in IL-10 intervention group (group I). Hepatic fibrosis was induced in rats, in the 7~(th) week And the 11~(th) week the hepatic stellate cells were isolated from the rats in these three groups by means of sequential perfusion with collagenase and pronase E and elutriation over a 11% Nycodenz gradient. The expression of ICAM-1 in the hepatic stellate cells were detected by semiquantitative reverse transcription polymerase chain reaction. RESULTSThe hepatic fibrosis model was established and the hepatic stellate cells were isolated sucessfully. ICAM-1 was detected in the group C and group I but not in the group N, its level in the group C was higher than that in the group I (P(0.01)). In the group C the expression of ICAM-1 in the 11~(th) week was higher than that in the 7~(th) week (P(0.01)), and in the group I the expression was decreased significantly (P(0.01)) compared to that in the group C in the 11~(th) week. CONCLUSIONThe expression of ICAM-1 in hepatic stellate cells is up-regulated during development of experimental hepatic fibrosis, and exogenic IL-10 may inhibit this response so as to produce antifibrogenesis effect.
