The Effect of Anti-vascular Endothelial Growth Factor Monoclonal Antibody in Alleviating Hepatic Ischemia/reperfusion Injury

Abstract

Objective To investigate the effect of anti-vascular endothelial growth factor monoclonal antibody(anti-VEGFmAb) in alleviating hepatic ischemia/reper-Fusion(I/R) injury.Methods Thirty normal male Sprague-Dawley rats were randomly divided into three groups,namely group A which were subjected to I/R injury with intraperitoneal administration of anti-VEGF mAb(100 μg/300 g) 15min before ischemia;group B with the same injury followed by saline administration in the same manner,and group C with only anesthetization and leparotomy but not ischemia.All rats were killed at 6 hours after reperfusion.Routine assays were performed for testing the levels of alanine aminotransferase(ALT)、aspartate aminotransferase(AST)、lactate dehydrogenase(LDH)、the activity of hepatic myeloperoxidase(MPO) and the cell apoptosis.The expression of VEGF in the liver was used to the examination by means of Western blot and the pathological changes in the liver were evaluated with hematoxylin and eosin(HE) stained sections.Results The levels of serum ALT,AST,LDH and MPO activity in hepatic tissues increased after hepatic ischemia/reperfusion injury(all P0.01),and expression of VEGF protein was also increased significantly.But these effects were offset by administration of anti-VEGF mAb(P0.01).In group B,widespread pathological changes and cell apoptosis were observed in the hepatic tissue following hepatic ischemia/reperfusion injury,while similar changes were scarcely visible in groupe A due to the protective effect of intraperitoneal administration of anti-VEGF mAb 15min before ischemia.Conclusion Hepatic ischemia/reperfusion injury can be alleviated by anti-VEGF mAb treatment.
