Effects of FHIT Gene Transfection on Proliferation and Invasion of Human Colon Cancer Cell Line SW480

Abstract

Objective To investigate the effects of FHIT gene transfection on proliferation and invasion of human colon cancer cell line sw480.Methods Eukaryotic expression plasmid containing FHIT,pRc/CMV2-FHIT,was introduced into E.coli DH5α and amplified.The plasmid was purified and identified by restrictive enzyme digestion.Plasmid pRc/CMV2-FHIT was transfected into SW480 cells,and positive cell clones(SW480-FHIT)were selected and amplified.plasmid-transfected SW480 cells(SW480-pRc/CMV2)and normal SW480 cells were set as control.Reverse transcription-polymerase chain reaction(RT-PCR)was applied to assess the expression of FHIT.Proliferation and invasion ability of SW480 cells were measured before and after transfection by MTT assay and cell invasion assay,respectively.Results SW480 cell transfected with pRc/CWV2-FHIT was approve to express FHIT,while FHIT expression was completely absent in the control group.The expression of FHIT significantly inhibited the proliferation and the invasion of SW480 cells in the experimental group(P0.01).Conclusion Re-expression of FHIT gene using gene transfection technology effectively inhibits the growth and proliferation of SW480 cell,and induces apoptosis of SW480 cell.The pRc/CMV2-FHIT rovides novel therapeutic potentials for the treatment of human colon carcinoma.
