Toxic Effect of Thrombin to Prihematomal Tissue Following Intracerebral Hemorrhage

Abstract

Aim:To investigate the toxic effect of thrombin on perihematomal tissue injuries in intracerebral hemorrhage (ICH).Methods:Ninety rats were divided into five groups:ICH groups,ICH plus hirudin groups,thrombin groups,thrombin plus hirdin groups,and sham operation groups.48 h later,all rats were decapitated after neurological deficit scores of rats were evaluated,then brain water content and MPO activities were measured and the expression of TUNEL positive cells in perhimatoma were detected.Results:Compared with ICH group,TNEL positive cells,brain water content,and MPO activities in perihematoma were all decreased significantly when recombinant hirudin was injected immediately into hematoma (P0.01),so did the NDS of rats.When 10 U thrombin was injected into the right caudate nucleus,there were higher content of brain water and MPO activities in local tissue,a number of TUNEL positive cells were detected as well.Conclusion:Thrombin following ICH may be involved in preihematomal edema,leukocuyte infiltration and neuronal DNA damage.
