The Influence of Different Dyeing Methods on Pigeon Hippocampal Neuron Forms

Abstract

The frozen sections from pigeons' brains were stained with haematoxylin-eosin( HE),methylene blue,cresyl violet,neutral red and Cajal's staining method respectively to explore the influence of different dyeing methods on pigeon hippocampal neuron forms,then the shape and size of neurons were compared and analyzed under optical microscope. It is found that perikaryons,axons,dendrites and nucleus are more clearly seen when stained with cresyl violet; The length and diameter in the neurons stained with neutral red are greater than that in the methylene blue( p 0. 05) and cresyl violet staining( p 0. 01); and perikaryons and necleus stained with Cajal's staining method are smaller( p 0. 01) than that of other methods. It is concluded that different staining methods have influence on showing the shape and size of neurons,and the staining method with cresyl violet is the best way to show the histological structure of the neurons in the hippocampus of pigeons.
