Abstract

AIM To observe the preventive effect of zafirlukast against acute lung injury. METHODS 24 New-Zealand rabbits were randomly divided into three groups: Control group (n=8), injury group (n=8), and treatment group (n=8). into rats in injury group was Endotoxin (LPS, 1 mL·kg -1), normal saline (1 mL·kg -1) in control group, zafirlukast in treatment group (20 mg·kg -1) twice before being injected endotoxin (LPS, 1 mL·kg -1) half an hour and one hour. Observe the arterial oxygen pressure (PaO 2), the pulmonary arterial pressure, count of peripheral-blood leukocytes, NO concentration in the BALF, lung index (wet weight/body weight), lung's wet /dry weight ratio (W/D) and had HE staining of the pathological sections were observed respectively. RESULTS The pulmonary arterial pressure and NO concentration in treatment group had no significant difference with that in injury group (P0.05), PaO 2 in treatment group significantly improved (P0.01). Lung index and W/D ratio in treatment group were significantly lower than those in injury group (P0.01), the count of peripheral-blood leukocytes in treatment group was higher than that in injury group (P0.01). HE staining showed that lung injury in treatment group was improved. CONCLUSION Zafirlukast has preventive effect against acute lung injury.
