Abstract
AIM To investigate the effect of sodium selenite on viral protein synthesis and hepatitis B virus(HBV)DNA replication in vitro.METHODS The HBV DNA transfected cell line HepG2.2.15 was treated with different concentrations of sodium selenite for 3 days.Hepatitis B surface antigen(HBsAg),hepatitis B e antigen(HBeAg),Hepatitis B core antigen(HBcAg)and level of HBV DNA in supernatant were detected by enzyme-linked immunosorbent assay(ELISA).Changes of p53 were detected by immunohistochemical method in cell line HepG2.2.15.RESULTS Sodium selenite could suppress the expression of HBsAg and HBeAg,and inhibit the HBV DNA replication in a dose-dependent manner.The suppression rate of HBsAg was lesser than that of HBeAg.HBcAg under the action of sodium selenite showed similar effect on HBV DNA replication(P0.01),with the expression change of p53.CONLUSION Sodium selenite can inhibit the expressions of HBsAg, HBeAg,HBcAg in the HepG2.2.15 all line and HBV DNA replication through interference with the possible mechanism of abnormal expression of p53 protein.
