Abstract
Objective To investigate the isolation,culture,proliferation and differentiation of endothelial progenitor cells(EPCs) from human circulating blood in vitro.Methods The mononuclear cells were obtained by Ficoll density gradient centrifugation.The isolated cells were suspended in Medium 199 with 20% fetal blood serum(FBS) and vascular endothelial growth factor(VEGF) for culturing,differentiating and proliferating.The expressions of specific antigens on cell surface were analysed by immunohistochemistry,immunocytochemistry and fluorescence-activated cell sorting(FACS).Results The culture cells were positively expressed for vwF,CD31 and CD34 and could be shown to endocytose ac-LDL and UEA-I.Conclusion EPCs can be obtained from mononuclear cells in peripheral blood and be differentiated into endothelial cells in vitro.
