Abstract

Objective To investigate the effects of a new houttuyfonate derivative(HD) on proliferation of VSMCs and expression of Syndecan-4 induced by TNF-α in vitro.Methods Rat VSMCs were cultured and exposed to TNF-α or HD respectively,and then cotreated with TNF-α and HD.All the groups were cultured for 24 hours in vitro,in addition to the untreated control group established for comparison.The ratio of proliferation of VSMCs was determined by non-radioactive MTS/PMS assay and the expression of Syndecan-4 was evaluated by western blot using anti-Syndecan-4 antibody.Results Statistical analysis showed that,compared to the control group,HD alone had no effect on VSMCs growth,but significantly inhibited VSMCs proliferation induced by TNF-α.It also showed that compared to control group,HD alone had no effect on the expression of Syndecan-4,but significantly inhibited the expression induced by TNF-α(P0.05).Conclusions HD can inhibit proliferation of rat VSMCs and expression of syndean-4 protein induced by TNF-α in vitro.
