R848 synergized with CD3 antibody induces spleen lymphocyte functional changes in C57BL/6 mice

Abstract
This study performed to detect and analysis the roles of R848 on synergy with anti-CD3 in inducing spleen lymphocytes functional changes in C57BL/6 mice. Lymphocytes were isolated from spleens of normal C57BL/6 mice and cultured with medium, anti-CD3, R848 or anti-CD3 +R848 for 72 h. Then, MTT experiment was adopted to analyze the proliferation of lymphocytes; the level of IFN-γ and IL-4 in the supernatants of cultured cells was measured by ELISA; the content of different lymph cell populations and intracellular cytokines(IFN-γ and IL-4) were detected by FACS. Results indicated that effects of R848 on lymphocyte proliferation were not obvious, but anti-CD3 plus R848 could promote anti-CD3 to induce lymphocyte proliferation. Also, R848 and anti-CD3 had synergistic effect on the production of IFN-γ and IL-4 by CD4+T and CD8+T cells. Furthermore, R848 could induce lymphocyte activation, especially B cells. In conclusion, R848 could assist the effects of antiCD3 of inducing spleen lymphocyte proliferation and IFN-γ and IL-4 secreting, and the mechanism might related to R848-induced B lymphocyte activation.
