Abstract
The orthogonal design, L_9 (3~4), was adopted with 3 factors and 3 levels: basic medium (A: 1/4 MS, MS and 1/2 MS), mercaptoethanol (B: 0.5, 0.1 and 0 g/L) and activated carbon (C: 2.5, 5 and 0 g/L) in tissue culture. Leaf-derived calli of pear variety quot;Xueqingquot; were used as explants and cultured under the conditions of 25±1℃, 2 000 lx and pH 5.8. Browning rate of the explants were recorded 30 days later. A_1B_1C_2 appeared to e the optimum treatment in which browning rate was decreased to approximately 5%. The factors B and C influenced explant browning significantly (Plt;0.05) and significant differences were observed among different levels of B and C.
