Abstract
Objective To investigate the effect of high glucose on the expressions of transforming growth factor(TGF-β1)and platelet derived growth factor(PDGF BB) in cultured rat lomerular mesangial cells. Methods The rat glomerular mesangial cells were cultured in vitro and were divided into three groups after the subculture,namely,the normal glucose group(5.6 mmol/L),the high glucose group A(15 glucose mmol/L) and the high glucose group B(30 glucose mmol/L).These groups were cultured for 12 hours,24 hours and 48 hours respectively.The expression levels of TGF-β1 and PDGF BB mRNA in cells of each group were measured by RT PCR and the protein expression of TGF-β1 and PDGF BB were detected by means of immunocytochemistry. Results ①The rat glomerular mesangial cells of the normal group all had the expressions of TGF-β1 and PDGF BB.②Compared with the normal glucose group,cells of both the high glucose groups had increasing expressions of TGF-β1,PDGF BB mRNA and protein(P0.05).③Compared with those of the high glucose group A,the high glucose group B cells had increasing expressions of TGF-β1,PDGF BB mRNA and protein(P0.05). Conclusion High glucose can increase the expressions of TGF-β1,PDGF-BB mRNA and protein in cultured rat glomerular mesangial cells.
