Effect of GDM on MMP-2 and MMP-9 activities in glioma cells induced by HGF

Abstract

Objective To investigate the effects of geldanamycin (GDM) on the matrix metalloproteinases (MMP)-2 and MMP-9 activities in glioma cell induced by hepatocyte growth factor (HGF).Methods U251-MG and U87-MG of human malignant glioma cells were cultured by cell culture technique.MMP-2 and MMP-9 activities were examined by photodensity zymogram methods.Results MMP-2 and MMP-9 activities in U251-MG and U87-MG cells treated with HGF for 24 h increased significantly compared with those of normal control group (P0.05),while GDM inhibited this increasing tendency obviously (P0.05).Also 1 000 nmol/L GDM significantly inhibited the increase of MMP-2 and MMP-9 activities in these two glioma cells induced by HGF(P0.05).Conclusions GDM could inhibit the increase of MMP-2 and MMP-9 activities in U251-MG and U87-MG cells,also significantly depress their activity increase in these two glioma cells induced by HGF.
