Abstract
Objective To investigate the expression of Bcl-2 and Cytochrome C in hippocampus of chronic stress depression rats and the interference of lithium carbonate.Methods The 45 rats were randomly divided into depression group,lithium carbonate group and control group,with each group 15 cases.The depression group and lithium carbonate group were both given 21 days stress,and meanwhile the control group was fed normally.The lithium carbonate group was also given intragastric administration of lithium carbonate(60 mg/kg),while the depression group and control group were given intragastric administration of 0.9% sodium chloride with the same volume.The ethology examination was performed by using open-field and experiment of fluid consumption.The expression of Bcl-2 and Cytochrome C was detected by Western-blot method.Results Before the chronic stress,the horizontal crossing numbers,erection times,modification times and the percentage of sacchar-consumption among the three groups showed no statistically significant difference(P0.05),but they showed statistically significant differences after stress(P0.05).After stress,the horizontal crossing numbers,erection times,modification times and percentage of sacchar-consumption in depression group and lithium carbonate group were significantly lower than the control group(P0.05).The horizontal crossing numbers,erection times,modification times and percentage of sacchar-consumption in lithium carbonate group were higher than the depression group,but showed no statistically significant difference(P0.05).According to Western-blot method,the expression of Bcl-2 and Cytochrome C among the three groups after chronic stress showed statistically significant differences(P0.05).The expression of Bcl-2 in depression group was significantly lower than the control group,but the expression of Cytochrome C was significantly higher than the control group(P0.01).The expression of Cytochrome C in lithium carbonate group was significantly higher than the control group(P0.01).The expression of Bcl-2 in lithium carbonate group was lower than the control group,but showed no statistically significant difference(P0.05).The expression of Bcl-2 in lithium carbonate group was significantly higher than the depression group,but the expression of Cytochrome C was significantly lower than the depression group(P0.01).Conclusion The expression of Bcl-2 in hippocampus of chronic stress depression rats is decreased and the expression of Cytochrome C is increased,and they both can be adjusted by lithium.
