Abstract

Objective:To explore the effect and molecular mechanism of Qidankeli in the treatment of pulmonary fibrosis.Methods:40 Wistar rats induced pulmonary fibrosis were divided into normal control、fibrosis、Qidan and corticosteroids groups.Normal control group was treated with 0.9% saline intratracheally (2ml/kg).Fibrosis group was treated with bleomycinA5(5mg/kg) intratracheally.Qidan group was given orally with Qidankeli (250mg/kg) daily.Corticosteroids group was treated with Hydrocortisone(25mg/kg,im) daily.All rats were killed 28 days later and sent to pathological examination.The hybridizations were used to analyze the expression of TGF-β1 mRNA、TNF-α mRNA in the lung tissue of rats.Results:There were more heavy alveolitis,pulmonary fibrosis and more collagen accumulation in fibrosis group,while the low-grade in Qidan group and middle-grade changes in corticosteroids group were observed.The demi-quantification image analysis of TGF-β1 mRNA in fibrosis group,corticosteroids group,Qidan group were(3.4320±0.2063),(1.8560±0.1356),(2.6920±0.2575),(1.0542±0.4288)mm~2.The demi-quantification image analysis of TNF-αmRNA in fibrosis group,corticosteroids group,Qidan group were (3.7500 ±0.1713),(1.9100±0.1935),(2、6220±0.3815),(1.480±0.1221)mm~2.The difference between fibrosis group and other groups,corticosteroids group and normal control group and Qidan [WTBZ]group were significant(P0.01).Conclusions:Qidankeli could decrease the expression of TGF-β1、TNF-α in the lung tissue of rats and obviously alleviate pulmonary fibrosis.
