Abstract

Objective To analyze arsenic Speciation and concentrations in urine of rats in different groups,to seek for different aspects of metabolism between sodium arsenate and sodium arsenite,and to understand further the basic data about relationship between it's metabolism and mechanism of toxicity.Methods Seventy rats were randomly divided into 7 groups such as control group,sodium arsenite high,middle and low dose group and sodium arsenate high,middle and low dose group.When 70 rat were fed with oral administration respectively deionized water,sodium arsenite and sodium arsenate for 1 months,the urine of every group were collected.Then appied the high efficiency liquid chromatography and hybrid genesis atomic fluorescence spectroscopy(HPLC-HGAFS) to determine Speciation and concentrations of sodium arsenate and sodium arsenite and it's product of metabolism in urine of rats.Meanwhile,the constituent ratio in different groups would be compared.Results The constituent ratio of iAs3+,iAs5+ and DMA were 36.1%∶3.0%∶60.8% in urines of rat in sodium arsenite middle groups,while the constituent ratio of iAs3+,iAs5+ and DMA were 55.9%∶0.8%∶43.4% in urine of rats in sodium arsenate middle groups,there were significant differences in constituent ratio between sodium arsenate and sodium arsenite(χ2=8.463,P0.05).For high and low groups,the significant different in constituent ratio were disappeared.Conclusion The internal metabolism is significantly different between sodium arsenate and sodium arsenite in urine.
