β-amyloid protein induced neuronal apoptosis in the brain

Abstract

Objective To investigate the role of apoptosis in pathogenesis of A β in brain. Methods Aβ 1 40 was injected into CA 1 subfield of rat right hippocampus.Seven days later,apoptosis was determined by using of HE staining、TUNEL staining and Transmission Election Microscopy (TEM),and the expression of Bax/Bcl 2 was determined with immunohistochemistry SABC method. Result In A β group,HE、TUNEL staining and TEM examination revealed many apoptosis. In the sham operation and normal saline control group,however,there was not the above mentioned changes.Comparision with the control groups,the expression of Bax in hippocampal CA 1 subfield in A β group was stronger,but the expression of Bcl 2 was no obvious difference between the three groups. Conclusion The result of this study indicated that A β can induce neuronal apoptosis in brain,and the stronger expression of Bax may play a important role in those neuronal apoptosis,which supports the idea that apoptosis take part in the pathogenesis of AD.
