Abstract
Objective:To investigate the effect of cisplatin on apoptosis and p53 protein expres- sion of children testicular yolk sac tumor,to find out the mechanism of its anticancer action. Methods: By techniques of cell culture in vitro, yolk sac tumor was treated with cisplatin. Then, the apoptosis index was measured with TUNEL technique and p53 protein expressions of the cells were observed with immunocytochemistry staining technique. Results:Under the effect of cisplatin, the round shrinkage of cells took place and the cancer cells desquamated from the wall. The apoptotic body was formed. The apoptosis index was increased with the increasing of the concentration and the prolonging of time. (P 0.01). p53 gene expression has reached the top level in early stage,thereafter,no lasting enhancement was found. Conclusion: Cisplatin can induce the apoptosis of the children's testicular yolk sac tumor cell, increase the expression of p53 gene protein, and the increasing of the expression of p53 gene may be one of an important mechanisms of cisplating's anticancer action.
