Study on Cell Suspension Culture for Leaf Mustard

Abstract

The cotyledons of leaf mustard were taken as explants to study the techniques of callus induction and cell suspension culture.The results showed that the optimal medium for callus induction was MS+0.3 mg/L NAA+1.0 mg/L 6-BA+0.1 mg/L 2,4-D,while the optimal formula for callus proliferation was MS+3.0 mg/L 6-BA+0.1 mg/L 2,4-D.Cell growth process in suspension culture exhibited an S-shaped curve and a logarithmic growth phase was reached after 2 to 10 days' culture.When the calli of suspension culture were transferred to an MS medium containing 1.0 mg/L NAA and 3.0 mg/L 6-BA and sub-cultured twice,shoots differentiated from the calli.Rooting plantlets were induced on MS with 0.1 mg/L NAA.
