Abstract

Objective:To research the anti-proliferation effect of extract of Rhizoma Atractylodis on A549 cells.Method:Rhizoma Atractylodis was extracted by 90% ethanol using ultrasonic method for 1h,then supernatant was collected by centrifugation;The proliferation activity of A549 inhibited by extract of Rhizoma Atractylodis was measured by MTT assay.The effect of extract of Rhizoma Atractylodis on cell cyclic was detected by flow cytometry;cyclin D1 protein expression was measured by Western Blot.Result: Extract of Rhizoma Atractylodis can effectively inhibit the proliferation of A549 in dosage and time dependent manner,the best inhibitory effect was appeared on 48h,the IC50 is about 77.12μg/ml;extract of Rhizoma Atractylodis can block cell cyclic in G0/G1 phase,the G1 phase was increased to 53.41% when A549 cells inhibited for 48h and effectively decrease cyclin D1 protein expression.Conclusion: Rhizoma Atractylodis can block cell cyclic in G0/G1 phase via decrease cyclin D1 protein to realize anti-proliferation effect.
