Abstract
Several factors of affecting extraction rate of polysaccharide from Plenrotus eryngii fruit body such as extraction temperature, extraction times and the solid-liquid ratio (W:V) were studied by uniform design test. The result showed that the extraction temperature was the primary factor to affect the polysaccharide extraction rate. The optimum extraction conditions were as follows: the ratio of sample to water(1:50),temperature (100℃) and time (240mi.). The extraction rate was 4.35% under the optimal extraction conditions. The crude polysaccharide was purified by ethanol sedimentation, dialysis, DE-52 chromatography. The purified Plenrotus eryngii polysaccharide was examined with ultraviolet spectral analysis without any protein and nuclear acid. the maximum absorbing peak was in 192 nm.
