Abstract
Objective To study the changes of vascular endothelial growth factor(VEGF) and soluble Eselectin(sE-selectin) after different duration of one lung-ventilation(OLV) in rats.Methods Thirty SD rats were randomly divided into 5 groups(each n = 6):control group received double lung-ventilation.Group l,2,3,and 4 received OLV for 1 /2 h,1 h,2 h,and 4 h,respectively,with tracheal catheter inserted into right lung.The levels of TNF-α and IL-8 in lung tissues were determined,and the levels of sE-selectin in serum and VEGF in serum and lung tissues were determined by sandwich enzyme-linked immunosorbent assay(ELISA).Ultrastructural changes of lung tissues were observed by electric mirror.Results The levels of TNF-α and IL-8 in lung tissues were gradually increased along with the time of OLV.The levels of sE-selectin in serum in Group 2,3,and 4 were significantly increased than that in control group(P 0.05).The level of VEGF in lung tissues in Group 4 was decreased as compared with that in control group(P 0.05),while that in serum was increased(P 0.05) The ultrastructural changes of lung tissues were lamellar body emptied,mitochondria damaged in Group l,2,3,and 4.Conclusion Long time OLV could induce acute lung injury,in which VEGF and sE-selectin play an important role.
